چکیده
Background: Optimization of co-cultivation parameters during Agrobacterium-mediated transformation to Oxalis corniculata evaluated were bacterial density, infection period, acetosyringone (AS) concentration and temperature. Five fold efficiency is achieved in transient gus gene expression after optimizing various parameters. The optimized conditions Agrobacterium tumefaciens growth phase A600nm 0.17, period 30 min, addition medium (400 μM) cocultivation temperature 22°C. Methods: (L.) used for transformation. Bacterial culture was added 50 ml liquid YEP with kanamycin rifampicin grown until reaching the (A600nm). density ranged from 0.56,0.80 1.34 OD present study. made solid MS consisted BAP 2 mg L-1 NAA 0.5 concentrations AS at 0, 50, 100, 200, 400,600 800 μM. Histochemical analysis carried out. Result: Higher resulted more efficiency, but also higher necrosis explants. Dilution suspension reduced explants 64% when process out μM). Our studies proved that among conditions, critical mediated

Agrobacterium tumefaciens-mediated transformation of yeast.
Agrobacterium tumefaciens transfers a piece of its Ti plasmid DNA (transferred DNA or T-DNA) into plant cells during crown gall tumorigenesis. A. tumefaciens can transfer its T-DNA to a wide variety of hosts, including both dicotyledonous and monocotyledonous plants. We show that the host range of A. tumefaciens can be extended to include Saccharomyces cerevisiae. Additionally, we demonstrate t...
متن کاملAgrobacterium tumefaciens- mediated Transformation of Plant Cells
Agrobacterium tumefaciens is a Gram-negative soil bacterium that causes plant tumours by transferring a portion of DNA from a resident ‘tumour inducing’ (Ti) plasmid into plant cells where it is integrated into a plant chromosome and expressed. This bacteria’s capacity for DNA transfer is the basis of most current plant genetic engineering and make it a model system for the study of pathogenic ...
متن کاملAgrobacterium tumefaciens mediated transformation of Agaricus bisporus
Agrobacterium tumefaciens is known to transfer parts of its tumor-inducing plasmid to filamentous fungi including Agaricus bisporus. We have used this transformation system to transform germinating basidiospores of A. bisporus. Analysis of hygromycin B-resistant clones showed that the T-DNA integrated at random sites and in multiple copies into the host genome. Furthermore we have been able to ...

Abiotic stress conditions can be imposed on plants through either poor water quality or adverse climate condition. These types of stress affect plant growth and development throughout their life cycle. Plants that are tolerant to drought are therefore valuable, and turf grasses have the potential to grow tolerant. Turf grasses provide ground cover and offer multiple benefits, from their aesthet...
متن کاملAgrobacterium Mediated Transformation of Maize (Zea mays L.)
Agrobacterium tumefaciens mediated transformation may offer a better alternative than the biolistic gun for genetic transformation of maize plants. This gene delivery system results in a greater proportion of stable, low-copy number transgenic events than does the biolistic gun, and is highly efficient. In the present work, we studied maize transformation using A. tumefaciens by identifying som...
